Doctoral position in Medical Science

Project title: The role of gut mucosal phages in cardiometabolic disease

The Department of Molecular and Clinical Medicine is located at the Institute of Medicine at the Faculty of Medicine at Sahlgrenska Academy, University of Gothenburg.

General information about being a doctoral student at the University of Gothenburg can be found on the university's doctoral student pages.
https://www.gu.se/en/doctoral-studies

Duties

The PhD education in Medical Science comprises carrying out a scientific project and completing at least 30 credits of courses at third-cycle level. The doctoral student must also write a scientific compilation thesis or monograph corresponding to at least 120 credits. For more information about third-cycle studies at Sahlgrenska Academy, see https://www.gu.se/en/sahlgrenska-akademin/doctoral-studies

Doctoral studies comprises four years of full-time study, and leads to a doctoral degree.

As part of your employment as a doctoral student, you may have departmental duties corresponding to up to 20 % of full-time employment, distributed throughout your study period, and result in a corresponding extension of the studies. Departmental duties usually consist of teaching at first- and second-cycle levels, but may also include research and administration.

Project description:
Phages - viruses that infect bacteria - are the most common biological entity on our planet, as well as in the human body. Still, the potential role of phages in health and disease is insufficiently understood.

The gut is the largest reservoir of both bacteria and phages. However, while bacterial density is highest in the luminal contents, phages are instead enriched in the mucus that covers and protects the epithelial cells. This is because the phages bind to glycans (sugar molecules) on mucin glycoproteins. When fiber intake is low, bacteria in the large intestine may turn to these glycans as an alternate source of nutrition, liberating the attached phages. Due to their small size, these phages could easily cross epithelial and endothelial barriers into the intestinal tissue and circulation. Certain phages have been reported to trigger inappropriate antiviral immune responses that might contribute to chronic inflammation and cardiometabolic diseases.

In this project, the doctoral student will investigate if disruption of the triangular relationship of phages, bacteria and gut mucosal cells partly explains the association of low fiber intake and cardiometabolic diseases. First, we will study how the composition of phages, their translocation across the gut barrier and antiviral immune responses differ in colon mucosa with normal vs. impaired mucus protection. Second, we will determine the effects of a fiber-based, dietary intervention designed to prevent excessive mucus degradation. Finally, we will develop a model to predict disruption of the gut mucosal barrier. This model will be evaluated in non-invasive samples from individuals with cardiometabolic diseases, such as fatty liver disease and atherosclerosis, and healthy controls persons.

The project will combine clinical cohorts, molecular biology techniques and advanced data analysis, providing a solid foundation in translational medical science.
