The Department of Molecular and Clinical Medicine, at the Institute of Medicine, conducts translational research primarily on metabolism, diabetes, obesity and cardiovascular disease in order to improve health and healthcare in the community. The overall goal is to create an excellent translational research environment of a very high international standard.

The Romeo laboratory studies the biological and genetic mechanisms underlying metabolic dysfunction-associated steatotic liver disease (MASLD) and its progression to liver fibrosis and cirrhosis. Our research combines human genetics with experimental models to identify disease mechanisms and therapeutic targets. We use state-of-the-art cell culture systems, liver organoids and molecular biology approaches to investigate pathways involved in liver injury and fibrosis.

Duties

The successful candidate will participate in an experimental research project investigating genes involved in susceptibility to liver fibrosis and cirrhosis. The work will involve establishing and maintaining spheroid and organoid cultures derived from liver cell lines and primary human hepatocytes, as well as generating and maintaining stable mammalian cell lines. The successful candidate will also use a range of molecular biology techniques, including DNA and RNA isolation, DNA cloning, transfection, PCR, protein expression analyses, and RNA expression profiling.

In addition, the role includes carrying out biochemical techniques such as immunoblotting (Western blotting), immunoprecipitation, subcellular fractionation, and siRNA-mediated gene silencing. The project further involves conducting metabolic flux experiments using radioactive tracers, as well as immunocytochemistry and in situ hybridisation experiments. R- and Linux-based computational tools will be used to analyse genetic and genomic datasets.

The work includes documenting, analysing, and presenting experimental results and actively participating in regular laboratory meetings and scientific discussions. The position provides an opportunity to gain practical experience in translational liver research and experimental disease modelling.

Qualifications

The successful applicant should have a background in biomedicine, molecular biology, biotechnology, cell biology, or another relevant life science discipline. Previous experience with organoid and/or spheroid culture, or liver biology, is required, as is practical experience in molecular biology. A good command of spoken and written English is also essential for the position.

Experience in omics analysis is considered meritorious, as is experience with mammalian cell culture, including spheroids and/or organoids and the generation of stable cell lines. Experience with molecular biology techniques such as DNA and RNA extraction, cloning, PCR, transfection, and gene expression analysis will be considered an advantage. Similarly, experience with biochemical techniques, including Western blotting, immunoprecipitation, and immunocytochemistry, is desirable. Experience with RNA interference (siRNA) and the analysis of biological data using R and/or Linux-based environments is also considered meritorious. Previous experience in liver biology, metabolism, or experimental disease models will be viewed favorably.

Great emphasis will be placed on personal suitability. The successful applicant should be motivated, well organized, able to work both independently and as part of a research team and have a genuine interest in experimental biomedical research.
